Agomelatine is a melatonin receptor agonist (MT1 and MT2) as well as a 5-hydroxytryptamine receptor (5-HT2C) antagonist. Resynchronization of circadian rhythms, augmentation of dopaminergic and adrenergic input to the frontal cortex, induction of neurogenesis, and other processes all function in a complementary and possibly synergistic manner to ameliorate depressed states. Additionally, melatonergic agonism and 5-HT2C receptor antagonism work together to improve anxiety symptoms, sleep, and sexual function.
