Precautions and warnings

Half of the patients who are successfully treated will develop new gallstones within 10 years. This can also happen during breastfeeding (lactation).

Drug Interactions

Cholestyramine, charcoal, and antacids can make this medicine less effective. Antacids that contain aluminum can lower how much of the medicine your body absorbs. Estrogens and clofibrate may reduce how well ursodeoxycholic acid works by making your body release more cholesterol into the bile. This medicine might also increase the amount of ciclosporin in your blood. It could make dapsone less effective and may also lower the levels of ciprofloxacin and nitrendipine in your body.

Contraindications

Gallstones can be hard (calcified) or dark (pigment), and some can be seen on X-rays (radio-opaque). Problems also include gall bladders that don’t work properly, long-term liver disease, and peptic ulcers. Other concerns are inflammation in the colon or small intestine, as well as pregnancy.

Mode of actions

Ursodeoxycholic acid lowers the amount of cholesterol made and released by the liver, and it also reduces the absorption of cholesterol in the intestines.

Dosage & Administration

Adult: PO Dissolution of cholesterol-rich gallstones 6-12 mg/kg/day as a single dose at bedtime after radiological disappearance of stones. Max: 15 mg/kg.

Primary biliary cirrhosis 10-15 mg/kg/day in 2-4 divided doses. Prevention of gallstones in patients undergoing rapid wt loss 300 mg twice daily.

Pregnancy & Lactation

Although animal reproduction studies have shown an adverse effect even sometimes unable to demonstrate the risk on the fetus and there are no adequate and well-controlled studies in pregnant women, but during pregnancy, this medicine can be used just when definitely required.
