Precautions and warnings

Side-effects

Drug Interactions
將西洛他唑與其他影響酵素 CYP3A4（例如紅黴素、其他大環內酯類抗生素和地爾硫卓）或 CYP2C19（例如奧美拉唑）的藥物一起服用可以改變西洛他唑在體內的加工方式。
Contraindications
心臟衰竭;有出血傾向；有心律異常病史；QT間期長；嚴重的腎臟問題；中度至重度肝臟問題。
Mode of actions
西洛他唑是一種以選擇性和可逆方式阻斷磷酸二酯酶-III (PDE-III) 酶的藥物。 透過這樣做，它可以阻止體內一種稱為環磷酸腺苷 (cAMP) 的物質的分解。 血小板和血管中較高水平的 cAMP 可以防止血小板聚集在一起，並幫助血管放鬆和擴張。 此外，西洛他唑可阻止細胞攝取腺苷，從而因其對 PDE-III 的影響而進一步提高 cAMP 水平。
Dosage & Administration
成人：PO 100 mg，每日兩次。
Pregnancy & Lactation
雖然動物生殖研究顯示對胎兒有不良影響，並且在孕婦中沒有充分且良好對照的研究，但在懷孕期間，儘管存在潛在風險，但在確實需要時可以使用該藥物。
在懷孕最後 3 個月內使用這種藥物的新生兒母親可能很少會出現困倦、肌肉僵硬或顫抖、餵食或呼吸困難、持續哭泣等症狀。 如果您發現寶寶有任何這些症狀，尤其是在第一個月內，請立即告訴醫生。
