Precautions and warnings
Des saignements intermenstruels peuvent survenir chez les personnes prenant un traitement contre l'endométriose. Ce traitement peut également provoquer une certaine rétention d'eau, c'est pourquoi les patients souffrant d'épilepsie, de migraine, d'asthme, de problèmes cardiaques ou rénaux doivent être surveillés attentivement. Certaines personnes peuvent également avoir une tolérance réduite au glucose. Les personnes souffrant de dépression, de diabète, d'épilepsie, d'asthme, de migraine, d'hypertension artérielle ou de problèmes rénaux ou cardiaques doivent être étroitement surveillées. Surveillez les signes de changements de vision, de problèmes oculaires et de caillots sanguins. Soyez prudent si la patiente allaite.

Drug Interactions
L'aminoglutéthimide et certains médicaments qui augmentent l'activité enzymatique (tels que la carbamazépine, la griséofulvine, le phénobarbital, la rifampicine et la phénytoïne) peuvent réduire la quantité de médicament dans le sang, le rendant ainsi moins efficace. Si la médroxyprogestérone est utilisée à des fins contraceptives avec ces médicaments, des précautions supplémentaires ou des méthodes contraceptives supplémentaires peuvent être nécessaires.

Mode of actions
La médroxyprogestérone est une hormone artificielle semblable à la progestérone. Il modifie la muqueuse de l'utérus de la phase de croissance à la phase de sécrétion. Bien qu’il ait des actions faibles similaires à celles des hormones mâles (androgènes) et favorise la construction des tissus (effets anabolisants), il n’agit pas comme les œstrogènes. Lorsqu'il est administré par injection, il bloque la libération de certaines hormones par l'hypophyse, ce qui empêche les ovules de mûrir et d'être libérés par les ovaires.
Dosage & Administration
Adulte : PO Ménorragie 2,5 à 10 mg/jour pendant 5 à 10 jours les jours 16 à 21 du cycle menstruel. Répétez l'opération pendant 2 cycles.
Endométriose légère à modérée 10 mg 3 fois/jour pendant 90 jours consécutifs, à compter du premier jour du cycle menstruel.
En tant que composant progestatif du THS pour la ménopause, la posologie dépend de la composante œstrogène du traitement. Plusieurs schémas thérapeutiques sont utilisés : 1,5 mg, 2,5 mg ou 5 mg/jour ; 5 ou 10 mg/jour pendant 12 à 14 jours sur un cycle de 28 jours ; 20 mg/jour pendant 14 jours sur un cycle de 91 jours.
Saignements utérins anormaux dus à un déséquilibre hormonal en l'absence de pathologie organique : À partir du 16ème ou du 21ème jour calculé du cycle menstruel, 5 ou 10 mg d'acétate de médroxyprogestérone peuvent être administrés quotidiennement pendant 5 à 10 jours.
Réduction de l'hyperplasie de l'endomètre chez les femmes ménopausées recevant quotidiennement 0,625 mg d'œstrogènes conjugués : Les comprimés d'acétate de médroxyprogestérone peuvent être administrés à des doses de 5 ou 10 mg par jour pendant 12 à 14 jours consécutifs par mois, chez les femmes ménopausées recevant quotidiennement 0,625 mg d'œstrogènes conjugués, soit à partir du 1er jour du cycle, soit du 16e jour du cycle.
Cancer du sein 0,4-1,5 g/jour. Max : 2 g/jour.
Palliation du carcinome de l'endomètre et du rein 200-600 mg/jour.
Palliation du carcinome de la prostate 100-600 mg/jour.
Aménorrhée secondaire 5 à 10 mg/jour pendant 5 à 10 jours. Répétez pendant 3 cycles.
Pregnancy & Lactation
Ce médicament ne doit pas être utilisé pendant la grossesse. Il peut être absorbé par la peau et les poumons, ce qui peut endommager les organes de votre enfant à naître et provoquer des handicaps congénitaux. Deux types stricts de contraception efficaces en commençant un mois avant et pendant le traitement et pendant trois ans après l'arrêt de ce médicament. Discutez avec vos médecins si vous ne savez pas quels types de contraception sont efficaces pour vous. Vous devez passer des tests de grossesse régulièrement pendant votre traitement par ce médicament et pendant au moins trois ans après la prise de ce médicament. S'il semble que vous devenez enceinte, arrêtez de prendre ce médicament et appelez immédiatement votre médecin. Si vous tombez réellement enceinte, manquez vos règles ou avez des relations sexuelles sans utiliser deux formes de contraception. Dans certains cas, votre médecin peut vous prescrire une contraception d’urgence pour éviter une grossesse. Les personnes prenant ce médicament ne doivent pas donner de sang pendant le traitement et pendant trois ans par la suite.
Ce médicament est également contre-indiqué dans le lait maternel. L'allaitement n'est pas recommandé pendant l'utilisation de ce médicament et pendant trois ans par la suite.
