Precautions and warnings
Perdarahan terobosan dapat terjadi pada orang yang menjalani pengobatan endometriosis. Perawatan ini juga dapat menyebabkan retensi cairan, sehingga pasien dengan kondisi seperti epilepsi, migrain, asma, masalah jantung, atau ginjal harus diawasi dengan cermat. Beberapa orang mungkin juga mengalami penurunan toleransi glukosa. Orang dengan depresi, diabetes, epilepsi, asma, migrain, tekanan darah tinggi, atau masalah ginjal atau jantung harus diawasi secara ketat. Perhatikan tanda-tanda perubahan penglihatan, masalah mata, dan pembekuan darah. Berhati-hatilah jika pasien sedang menyusui.

Drug Interactions
Aminoglutethimide dan obat-obatan tertentu yang meningkatkan aktivitas enzim (seperti karbamazepin, griseofulvin, fenobarbital, rifampisin, dan fenitoin) dapat menurunkan jumlah obat dalam darah sehingga kurang efektif. Jika medroksiprogesteron digunakan untuk pengendalian kelahiran bersama dengan obat-obatan ini, tindakan pencegahan ekstra atau metode pengendalian kelahiran tambahan mungkin diperlukan.

Mode of actions
Medroksiprogesteron adalah hormon buatan yang mirip dengan progesteron. Ini mengubah lapisan rahim dari fase pertumbuhan ke fase sekresi. Meskipun ia memiliki beberapa tindakan lemah yang mirip dengan hormon pria (androgen) dan mendorong pembentukan jaringan (efek anabolik), ia tidak bertindak seperti estrogen. Ketika diberikan melalui suntikan, obat ini menghalangi pelepasan hormon tertentu dari kelenjar pituitari, yang menghentikan pematangan sel telur dan pelepasannya dari ovarium.
Dosage & Administration
Dewasa: PO Menoragia 2,5-10 mg/hari selama 5-10 hari pada hari ke 16-21 siklus menstruasi. Ulangi selama 2 siklus.
Endometriosis ringan sampai sedang 10 mg 3 kali/hari selama 90 hari berturut-turut, dimulai pada hari pertama siklus menstruasi.
Sebagai komponen progestogen pada HRT menopause Dosis tergantung pada komponen estrogen dalam terapi, beberapa regimen yang digunakan: 1,5 mg, 2,5 mg atau 5 mg/hari; 5 atau 10 mg/hari selama 12-14 hari dari siklus 28 hari; 20 mg/hari selama 14 hari dari siklus 91 hari.
Perdarahan uterus abnormal karena ketidakseimbangan hormonal tanpa adanya patologi organik: Dimulai pada hari ke-16 atau ke-21 dari siklus menstruasi, 5 atau 10 mg Medroksiprogesteron asetat dapat diberikan setiap hari selama 5 hingga 10 hari.
Pengurangan hiperplasia endometrium pada wanita pascamenopause yang menerima estrogen terkonjugasi 0,625 mg setiap hari : Tablet medroksiprogesteron asetat dapat diberikan dalam dosis 5 atau 10 mg setiap hari selama 12 hingga 14 hari berturut-turut per bulan, pada wanita pascamenopause yang menerima estrogen terkonjugasi 0,625 mg setiap hari, baik dimulai pada hari pertama siklus atau hari ke-16 siklus.
Kanker payudara 0,4-1,5 g/hari. Maks: 2 gram/hari.
Paliasi karsinoma endometrium dan ginjal 200-600 mg/hari.
Paliasi karsinoma prostat 100-600 mg/hari.
Amenore sekunder 5-10 mg/hari selama 5-10 hari. Ulangi selama 3 siklus.
Pregnancy & Lactation
Obat ini tidak boleh digunakan selama kehamilan. Dapat diserap melalui kulit dan paru-paru sehingga dapat merusak organ tubuh anak Anda yang belum lahir dan menyebabkan cacat bawaan. Ada dua jenis alat kontrasepsi yang efektif yang dimulai satu bulan sebelum dan selama pengobatan dan selama tiga tahun setelah menghentikan obat ini. Diskusikan dengan dokter Anda jika Anda tidak yakin jenis alat kontrasepsi mana yang efektif untuk Anda. Anda harus melakukan tes kehamilan secara rutin selama pengobatan dengan obat ini dan setidaknya tiga tahun setelah meminum obat ini. Jika tampaknya Anda hamil, hentikan penggunaan obat ini dan segera hubungi dokter. Jika Anda benar-benar hamil, melewatkan masa menstruasi, atau berhubungan seks tanpa menggunakan dua alat kontrasepsi. Dalam beberapa kasus, dokter Anda mungkin meresepkan kontrasepsi darurat untuk mencegah kehamilan. Orang yang menggunakan obat ini tidak boleh mendonorkan darahnya selama pengobatan dan tiga tahun setelahnya.
Obat ini juga dikontraindikasikan pada ASI. Menyusui tidak dianjurkan saat menggunakan obat ini dan selama tiga tahun setelahnya.
