Precautions and warnings

Patients with cardiovascular or liver illness, renal impairment, or recurrent eczema. Elderly. The medicine should be gradually tapered off.

Drug Interactions

Antimuscarinic and levodopa side effects are amplified. CNS stimulants are medicines that elevate the pH of the urine.

The use of Amantadine hydrochloride in combination with the intranasal administration of the live attenuated influenza vaccine (LAIV) has not been studied.

Contraindications

Hypersensitivity. Lactation and pregnancy. Seizures, severe renal impairment, and stomach ulcers are all symptoms of epilepsy or other seizure disorders.

Mode of actions

Amantadine is an antimuscarinic that is a mild dopamine agonist. It affects dopamine reuptake and release. It also has a noncompetitive antagonistic relationship with N-methyl-D-aspartate. It works as an antiviral medication to stop the influenza A virus from replicating.

Dosage & Administration

Adult: PO Influenza A 100 mg/day for 5 days. Prophylaxis of influenza A 100 mg/day for up to 6 wk. W/ influenza vaccination: up to 3 wk after vaccination.
Elderly: >65 yr: Daily dose of 1 day.
Child: 10-15 yr: 100 mg daily.

Herpes zoster 100 mg twice daily for 14 days, may continue for another 14 days if pain persists.
Parkinson's disease100 mg/day, up to 100 mg twice daily 1 wk or more. Max: 400 mg/day.

Renal impairment:
CrCl (ml/min)
>35 100 mg daily.
15-35 100 mg every 2-3 days.

Pregnancy & Lactation

Although animal reproduction studies have shown an adverse effect on the fetus and there are no adequate and well-controlled studies in pregnant women, but during pregnancy, this medicine can be used just when definitely required despite potential risks.

Mothers with a newborn baby who have used this drug during the last 3 months of pregnancy may seldom create symptoms including drowsiness, muscle stiffness or shakiness, feeding or breathing troubles, constant crying. If you notice any of these symptoms of your baby's particularly during their first month, tell the doctor immediately.

This medicine passes into breast milk. Talk to your doctor before breastfeeding.
