Precautions and warnings
مرگی کے شکار افراد، بوڑھے افراد اور ایسے کام کرنے والوں میں احتیاط برتیں جن کو ذہنی چوکنا رہنے کی ضرورت ہے۔ زاویہ بند ہونے والے گلوکوما، معدے یا آنتوں میں رکاوٹ، پیشاب کی نالی میں رکاوٹ، اوور ایکٹیو تھائیرائیڈ (ہائپر تھائیرائیڈزم)، آنکھ کے اندر دباؤ بڑھنا، دل کی بیماری، شدید دمہ، اور حمل کے دوران جیسے حالات میں بھی محتاط رہیں۔
Side-effects
- سی این ایس ڈپریشن، چکر آنا، سر درد، مسکن دوا؛ بچوں میں متضاد محرک؛ منہ کا خشک ہونا، سانس کی رطوبت گاڑھی ہونا، بینائی کا دھندلا پن، پیشاب کی روک تھام؛ جی آئی کی خرابی؛ خون کی خرابی.
Drug Interactions
امینوگلیکوسائیڈ اینٹی بائیوٹکس کی وجہ سے کان کے نقصان دہ اثرات (اوٹوٹوکسیسیٹی) کو ماسک کرتا ہے۔ لیووڈوپا کو معدے میں تیزی سے توڑ کر اور پیٹ کے خالی ہونے میں تاخیر کرکے اس کے جذب کو کم کرکے کم موثر بناتا ہے۔ cholinergic ادویات، جیسے tacrine اور donpezil، کے ساتھ ساتھ بعض antipsychotics کی تاثیر کو کم کرتا ہے۔ ہربل سپلیمنٹس جیسے ویلیرین، سینٹ جانز وورٹ، کاوا کاوا، اور گوٹو کولا غنودگی یا مسکن کو بڑھا سکتے ہیں۔
**خطرہ:**
الکحل، باربیٹیوریٹس، درد کش ادویات، نیند کی گولیاں اور اینٹی سائیکوٹکس کے ساتھ ملانا خطرناک طور پر مرکزی اعصابی نظام کی غنودگی یا افسردگی کو بڑھا سکتا ہے۔ جب MAOIs، atropine، یا tricyclic antidepressants (TCAs) کے ساتھ لیا جائے تو یہ antimuscarinic ضمنی اثرات کو بھی بڑھا سکتا ہے۔

Mode of actions
Diphenhydramine نظام ہضم، خون کی نالیوں اور ایئر ویز میں پائے جانے والے H1 ہسٹامائن ریسیپٹرز کو روک کر کام کرتا ہے۔ یہ آپ کو نیند کا احساس دلا سکتا ہے اور اس کے کچھ اثرات بھی دوائیوں سے ملتے جلتے ہیں جو acetylcholine (anticholinergic اثرات) کو روکتے ہیں۔
Dosage & Administration
زبانی
بالغ: ہائیڈروکلورائڈ کے طور پر: 25-50 ملی گرام روزانہ 3-4 بار۔ زیادہ سے زیادہ: 300 ملی گرام / دن۔
بچہ: 6.25-25 ملی گرام روزانہ 3-4 بار، تقسیم شدہ خوراکوں میں 5 ملی گرام/کلوگرام تک۔ زیادہ سے زیادہ: 300 ملی گرام / دن۔
حرکت کی بیماری کا علاج اور پروفیلیکسس
بالغ: بطور ڈیفین ہائیڈرمائن ڈی (ایسفیلینیٹ): معمول کی خوراک: 90-135 ملی گرام، اگر ضرورت ہو تو کم از کم 6 گھنٹے کے وقفوں پر دہرائی جا سکتی ہے۔ زیادہ سے زیادہ: 540 ملی گرام روزانہ۔ روک تھام کے لیے، سفر سے کم از کم 30 منٹ پہلے خوراک دی جائے۔
