Drug Interactions
Aminoglutethimide lowers the amount of tamoxifen in the blood.
Serious Warning: Taking aminoglutethimide with warfarin can make warfarin more effective, which increases the risk of dangerous bleeding. Using aminoglutethimide with other cancer drugs also raises the risk of blood clots.
Contraindications
Pregnancy and lactation. History of thromboembolic events.
Mode of actions
Tamoxifen is a type of drug called a triphenylethylene derivative. It works by attaching to estrogen receptors in tumors and other tissues, blocking estrogen from binding there. This reduces DNA production and stops the effects of estrogen. Instead of killing cancer cells directly, tamoxifen keeps them from growing by causing them to stay in the resting (G0) and first growth (G1) phases of the cell cycle.
Dosage & Administration
Adult: PO Breast cancer 20 mg in 1-2 divided doses. Max: 40 mg/day. Reduction of breast cancer incidence in high-risk women 20 mg/day for 5 yr.
Anovulatory infertility 20 mg/day on days 2-5 of the menstrual cycle. Max: 80 mg/day.

Pregnancy & Lactation
This medicine is not allowed during pregnancy. Do not use it if you are pregnant. It can be absorbed through the skin and lungs caused can damage your unborn child’s organs and cause congenital disabilities. Strict effective birth control during treatment and for six months after stopping this medicine. If you do become pregnant, stop taking this medicine and discuss your doctor right away.
There is no specific information on whether this medicine passes into breast milk or not. Considering the possible risks for the children, breastfeeding while using this medicine is not recommended and at least two weeks after the last dose. Consult your doctor before breastfeeding.
