Precautions and warnings
Do not use this medicine in people with psoriasis, blood or nervous system diseases, myasthenia gravis, liver or kidney problems, G6PD deficiency, epilepsy, or in children. It should also be avoided during pregnancy and breastfeeding. When giving this medicine by IV, infuse it slowly to reduce the risk of heart problems.

Drug Interactions
Taking phenylbutazone at the same time can make you more likely to get skin rashes, reduce how well neostigmine and pyridostigmine work, and lower how much ampicillin your body absorbs. Cimetidine can increase the amount of chloroquine in your blood by slowing its breakdown.
**Serious Risks:**
- When chloroquine is taken with halofantrine or other drugs that affect heart rhythm (like amiodarone), there is a higher chance of dangerous heart arrhythmias.
- There is also a greater risk of seizures when chloroquine is taken with mefloquine.
- Antacids can lower the absorption of chloroquine, so they should be taken at least 4 hours apart from each other.
- Rarely, a severe skin reaction (Stevens-Johnson syndrome) can occur if chloroquine is taken with pyrimethamine or sulphadoxine.
- Taking quinacrine with chloroquine can increase the risk of side effects.
Contraindications
Do not use if you have an allergy to the drug, possible or confirmed resistant P. falciparum infection, porphyria, retinal problems, or if you are taking other medications that are harmful to the liver.
Mode of actions
Chloroquine is used to prevent malaria and to treat sudden malaria attacks. It works well against the blood stages of *Plasmodium vivax*, *P. malariae*, and *P. falciparum*. The drug raises the pH inside the parasite's food vacuole, which disrupts the digestion of hemoglobin and affects how the parasite makes proteins. Chloroquine is also helpful in treating amoebiasis outside the intestines. For rheumatoid arthritis (RA), both chloroquine and the similar drug hydroxychloroquine can help slow down the progress of the disease—with hydroxychloroquine being more effective.
Dosage & Administration
Adult: PO Acute malaria As base: Initial: 600 mg, then 300 mg 6-8 hr later on day 1. Continue w/ 300 mg/day on days 2 and 3.
Malaria prophylaxis As base: 300 mg/wk, starting 1 wk before exposure, continue throughout on a wkly basis and for at least 4 wk after exposure.
Discoid and SLE; Rheumatoid arthritis As base: Initial: 150 mg/day. Max: 2.5 mg/kg/day.
Hepatic amoebiasis As base: W/ emetine or dehydroemetine: 600 mg/day for 2 days then 300 mg/day for 2-3 wk.
IV Severe malaria As base: 25 mg/kg given in several infusions over 30-32 hr.
Pregnancy & Lactation
Although animal reproduction studies have shown an adverse effect on the fetus and there are no adequate and well-controlled studies in pregnant women, but during pregnancy, this medicine can be used just when definitely required despite potential risks.
Mothers with a newborn baby who have used this drug during the last 3 months of pregnancy may seldom create symptoms including drowsiness, muscle stiffness or shakiness, feeding or breathing troubles, constant crying. If you notice any of these symptoms of your baby's particularly during their first month, tell the doctor immediately.
This medicine passes into breast milk. Talk to your doctor before breastfeeding.
