Check the patient’s left ventricular ejection fraction (LVEF) before starting treatment. Use caution in patients with moderate or severe liver problems. Be careful with patients who have, or are at risk of, a prolonged QTc interval, such as those with low potassium or magnesium levels. Monitor liver function and watch for lung symptoms that could suggest interstitial lung disease or pneumonitis. Use caution in pregnant or breastfeeding women.
The concentration of this drug increases if it is taken with CYP3A4 inhibitors (like ketoconazole, itraconazole, or grapefruit juice), as well as with midazolam, irinotecan, and digoxin. Its concentration decreases if used with CYP3A4 inducers (such as rifampin, carbamazepine, or phenytoin) or with proton-pump inhibitors (like esomeprazole). The drug’s behavior in the body can also change when taken with medicines that affect P-glycoprotein or BCRP (breast cancer resistance protein), or with drugs like topotecan and rosuvastatin. Taking it with paclitaxel or docetaxel can make side effects more likely and more severe. This medicine is processed mainly by the CYP3A4 or CYP2C8 enzymes.
Hypersensitivity to lapatinib ditosylate
This kinase inhibitor works by blocking the HER2 receptor (a type of EGF receptor) and its tyrosine kinase activity. By doing this, it may help stop the growth of new blood vessels (angiogenesis) and the multiplication of cancer cells.
HER2 positive metastatic breast cancer 1, 250 mg (5 tab) daily on days 1-21 continuously in combination w/ capecitabine. 2,000 mg/m2/day (orally in 2 doses approx 12 hr apart) on days 1-14 in a repeating 21-day cycle.
Hormone receptor positive, HER2 positive metastatic breast cancer Recommended dose: 1,500 mg (6 tab) continuously in combination w/ letrozole. All doses to be given once daily as a single dose.
This medicine is not allowed during pregnancy. Do not use it if you are pregnant. It can be absorbed through the skin and lungs caused can damage your unborn child’s organs and cause congenital disabilities. Strict effective birth control during treatment and for six months after stopping this medicine. If you do become pregnant, stop taking this medicine and discuss your doctor right away.
There is no specific information on whether this medicine passes into breast milk or not. Considering the possible risks for the children, breastfeeding while using this medicine is not recommended and at least two weeks after the last dose. Consult your doctor before breastfeeding.
