Amizide is indicated for

Precautions and warnings

Renal impairment is defined as a BUN level of 10 mmol/L and a serum creatinine level of 130 mmol/L. In extremely unwell patients, such as those with hepatic cirrhosis with ascites and metabolic alkalosis, or those with resistant oedema, serum electrolytes and blood urea levels should be monitored. Severe liver disease prior to admission. It's possible that parathyroid function will be affected. It's possible that you'll get hyperuricemia or gout. The levels of cholesterol and triglycerides may rise. Thiazides have the potential to alter glucose tolerance. DM can be worsened or triggered. It has the potential to activate or exacerbate SLE. Elderly. Hyponatraemia. Pregnancy.

Drug Interactions

Other antihypertensives have additive effects. It's possible that tubocurarine will make you more responsive. Reduces the response of the vascular system to pressor amines like norepinephrine. Alcohol, barbiturates, and opioids can cause orthostatic hypotension. To lessen the risk of first-dose hypotension, stop taking diuretics 2-3 days before starting an ACE inhibitor. Corticosteroids or ACTH can cause hypokalemia. NSAIDs reduce the diuretic, natriuretic, and antihypertensive effects of diuretics. Chlorpropamide causes increased hyponatraemia. Cyclosporin, tacrolimus, indometacin, and angiotensin II receptor antagonists all cause hyperkalemia.

Potentially Fatal: Increased risk of hyperkalaemia with other potassium-sparing diuretics (spironolactone or triamterene). Raise the risk of lithium toxicity.

Contraindications

Acute renal failure, severe progressive renal illness; severe hepatic failure, precoma linked with hepatic cirrhosis; Addison's disease; hypercalcemia; diabetic neuropathy; DM

Mode of actions

Amiloride is a mild natriuretic, diuretic, and antihypertensive antikaliuretic. These effects were found to be partially additive to the effects of thiazide diuretics in several clinical studies. Amiloride works by reducing Na reabsorption at the distal renal tubules, lowering the tubular lumen's net -ve potential, and lowering both K and hydrogen secretion, as well as their subsequent excretion. Hydrochlorothiazide is an antihypertensive and diuretic. It inhibits the reabsorption of electrolytes by the renal tubules. Hydrochlorothiazide raises Na and chloride excretion in roughly equal proportions. Natriuresis can cause a loss of potassium and bicarbonate. It exerts its hypotensive effect partly by reducing peripheral resistance.

Dosage & Administration

Adult: PO:
Hypertension Per tab contains amiloride 5 mg and hydrochlorothiazide 50 mg: 1/2 to 1 tab/day.

Congestive heart failure Per tab contains amiloride 5 mg and hydrochlorothiazide 50 mg: 1/2 to 2 tabs/day.

Hepatic cirrhosis with ascites and oedema Per tab contains amiloride 5 mg and hydrochlorothiazide 50 mg: 1-2 tabs/day.

Pregnancy & Lactation

Although animal reproduction studies have shown an adverse effect even sometimes unable to demonstrate the risk on the fetus and there are no adequate and well-controlled studies in pregnant women, but during pregnancy, this medicine can be used just when definitely required.
